Reasons Behind the Program Termination
Chief Executive Adrian Gottschalk noted that the compound successfully engaged its intended molecular target. The safety profile remained acceptable throughout testing, even when doses exceeded levels initially predicted by preclinical research.
However, clinical efficacy proved insufficient. Gottschalk explained that the underlying scientific rationale involving SMARCA2/4 interactions failed to produce therapeutic responses robust enough to justify continued investment.
This disappointing conclusion arrives after multiple years of collaborative development. The setback extends beyond a single experimental therapy.
Both organizations confirmed that a separate oncology collaboration focused on selective SMARCA2 degradation technology will also be discontinued. Foghorn indicated no additional cooperative initiatives with Lilly are anticipated moving forward.
The original collaboration was established in December 2021. Lilly’s Loxo Oncology division structured the agreement with $300 million in initial payments alongside an $80 million equity investment priced at $20 per share.
Those financial terms appeared favorable for Foghorn when announced. The current trading price stands dramatically below that initial $20 valuation.
Foghorn’s Path Forward
Following the partnership dissolution, Foghorn has implemented aggressive cost reduction strategies. The organization is eliminating nearly 40% of its workforce alongside broader operational restructuring.
Leadership believes these adjustments will extend available capital through the latter half of 2029. This timeline provides runway to sustain development of remaining pipeline candidates.
Foghorn stated it will concentrate resources on wholly-owned proprietary assets. This portfolio encompasses a selective EP300 degrader, a selective CBP degrader, and an oral program targeting immunology and inflammatory conditions.
The organization continues prioritizing its induced proximity technology platform as a foundational capability. These strategic areas operate independently of Lilly’s financial support.
For Eli Lilly, market reaction was minimal. Shares declined roughly 2%, an insignificant movement for a corporation of its scale.
For Foghorn, the implications are far more consequential. The biotech now operates primarily independently, relying on existing capital reserves and a streamlined organization to advance its development programs through the coming years.
